Vitamin B6 (Pyridoxine)

Summary

This marker assesses the genetic capacity of your microbiome to synthesize Vitamin B6 (Pyridoxine). This vitamin, in its active form (Pyridoxal 5'-Phosphate or PLP), is one of the most versatile "motors" in your biology, being the obligatory cofactor for over 100 different enzymes in your body. A high production capacity by your bacteria ensures continuous support for neurotransmitter synthesis, local immune system regulation, and amino acid metabolism.

Overview

Normally we get Vitamin B6 through our diet (meats, fish, chickpeas, potatoes). Your body absorbs it in the small intestine and sends it to the bloodstream, where it is used to build everything from red blood cells to brain chemical messengers.

However, the colon (large intestine) is an incredibly densely populated ecosystem that also needs Vitamin B6 to survive. This is where your producer bacteria come into play. Certain microbial species are true chemical factories capable of assembling the B6 molecule from scratch.

This bacterial B6 serves a dual purpose: on the one hand, it is partially absorbed by the cells of your colon for their own immune maintenance; on the other hand, it functions as a common good within the bacterial community. Many highly beneficial bacteria cannot produce B6 and depend absolutely on their neighbors to produce it in order to thrive and generate protective compounds like butyrate.

Health impact

The endogenous production of Vitamin B6 has a direct and bidirectional impact, connecting the health of your gut with that of your brain:

  • Gut-Brain Axis (Serotonin and GABA): B6 is the master switch for emotional well-being. Your body absolutely needs B6 to convert tryptophan into Serotonin (the happiness hormone) and glutamate into GABA (the calm neurotransmitter). A local and systemic deficit of this vitamin severely compromises the production of these molecules, affecting your mood, gut motility, and sleep quality.
  • Immune Inflammation Control: In the gut, a constant supply of bacterial B6 modulates the immune cells residing in the intestinal wall. It helps maintain a balance between cells that attack infections and cells that "calm" inflammation (regulatory T cells), preventing autoimmune reactions and allergies.
  • Cardiovascular Health: Together with B9 (Folate) and B12, Vitamin B6 is fundamental for recycling and neutralizing homocysteine, a toxic amino acid that, if it accumulates, severely damages the walls of your arteries and brain.

Technical report

The evaluation of de novo Vitamin B6 biosynthesis is performed by Shotgun metagenomic sequencing, quantifying the global genetic potential of the metabolic pathway in the ecosystem. Our bioinformatic algorithm tracks and maps the abundance of genes that make up the pyridoxal 5'-phosphate (PLP) biosynthesis superpathway, mainly the DXP-dependent route. It specifically searches for the load and potential expression of highly conserved key genes such as pdxS and pdxT (or pdxA/pdxJ depending on the alternative routes present in the clade).

At an ecological level, the ability to synthesize B6 is more widely distributed than that of other B vitamins, but its most efficient producers (primary synthesizers) usually belong to the phyla Bacteroidetes (e.g., species of Bacteroides and Prevotella) and certain classes of Proteobacteria. Important commensal bacteria such as many Firmicutes rely on cross-feeding to obtain their PLP.

High capacity indicates an autarkic and robust ecosystem. It ensures the health of the microbial trophic network and provides constant vitamin support for the host's neuroendocrine system. However, low capacity may indicate dietary dependence and ecological fragility. Low bacterial production of B6 can slow or stop butyrate synthesis by auxotrophic bacteria and compromise protein metabolism. If accompanied by symptoms of mental fatigue or sleep disorders, it is recommended to optimize dietary intake of B6, supplement with PLP (active form), and favor plant-based diets rich in complex polysaccharides to enhance the growth of the phylum Bacteroidetes.
