Vitamin B3 (Niacin)

Summary

This marker assesses the genetic capacity of your gut ecosystem to synthesize Vitamin B3 (Niacin). In the world of cell biology, niacin is the indispensable raw material for manufacturing NAD+, the "molecule of youth" and the universal energy currency of your cells. High bacterial production capacity ensures that your gut has the necessary energy to repair its own DNA, calm inflammation, and keep your entire microbial community functioning at peak efficiency.

Overview

Vitamin B3 or niacin is classically obtained through the diet (meats, fish, peanuts). Once in your body, it is converted into NAD+ (Nicotinamide Adenine Dinucleotide), a coenzyme without which life as we know it would stop in a matter of seconds, as it is responsible for extracting energy from food within your mitochondria.

However, the colon is an environment that consumes very large amounts of energy and is subjected to constant oxidative stress. Your intestinal cells cannot depend solely on the niacin that arrives through the blood.

Fortunately, a select group of gut bacteria possess the extraordinary ability to manufacture Vitamin B3 from scratch (from the amino acid tryptophan or aspartate). This bacterial niacin becomes a common good: it feeds the good bacteria that do not know how to manufacture it and, more importantly, is directly absorbed by your colon cells to recharge their local NAD+ levels.

Health impact

The endogenous production of Vitamin B3 has a profound and bidirectional impact on cellular longevity and immunological control:

  • DNA Repair and Anti-aging: The enzymes responsible for repairing DNA mutations (PARPs) and longevity proteins (Sirtuins) consume NAD+ massively to function. A constant bacterial supply of niacin protects colon cells from genetic damage, reducing the risk of polyps and premature tissue aging.
  • Intestinal Inflammation Brake: This is a fascinating clinical discovery. Niacin produced by bacteria directly binds to a specific receptor in the intestine called GPR109A (the same receptor that butyrate uses). By activating this switch, niacin "orders" immune cells to stop attacking and begin secreting anti-inflammatory substances (such as IL-10), acting as a vital shield against colitis and Crohn's disease.
  • Local Energy Metabolism: Without enough NAD+, your intestinal cells cannot use butyrate as fuel. Bacterial niacin ensures that your intestinal barrier's energy engine never shuts down.

Technical report

The evaluation of the de novo biosynthesis of niacin (Vitamin B3) is performed by Shotgun metagenomic sequencing, quantifying the global genetic potential of NAD synthesis pathways in the ecosystem. Our bioinformatics algorithm tracks and maps the abundance of genes and operons that make up the NAD biosynthesis superpathway (e.g., the aspartate pathway), specifically looking for the potential expression of highly conserved key genes such as nadA, nadB, nadC, nadD, and nadE.

At an ecological level, the main primary "producers" of B3 mostly belong to the phyla Bacteroidetes and certain Proteobacteria, as well as the species Akkermansia muciniphila. Paradoxically, the vast majority of bacteria from the phylum Firmicutes (including major butyrate producers like Faecalibacterium) are strictly niacin auxotrophs; that is, they depend on neighboring bacteria to supply it in order to survive.

High capacity indicates an autarkic and protective ecosystem. It guarantees the energy supply of beneficial auxotrophic bacteria and keeps the host's GPR109A receptor activated. Low capacity can signal an alert of ecological fragility and potential local oxidative stress. The gut depends 100% on systemic niacin. At a dietary level, adequate intake of tryptophan (niacin precursor) and vitamin B3 should be ensured, in addition to considering supplementation with forms (such as Nicotinamide Riboside - NR or NMN) to rapidly restore the NAD+ pool in the intestinal mucosa, combined with prebiotics to stimulate natural producers.
